How do I import CRISPR guides that have been generated by CRISPOR?
CRISPOR (crispor.org) is a web-based program that helps design, evaluate and clone guide sequences for the CRISPR/Cas9 system. If you use CRISPOR to design guides then you can export the results in Excel format and import them onto the same sequence in SnapGene.
CRISPOR calculates off-site (CFD and MIT) scoring for sites with a maximum of 4 mismatches. The SnapGene CRISPR tool currently calculates off-site scoring for sites with a maximum of 3 mismatches. This means that for the same target sequence and off-site genome, SnapGene may report higher off-sites scores compared to CRISPOR.
Copy the Sequence
Open and view the target sequence in SnapGene then go Edit → Select All and Edit → copy to copy the sequence to the clipboard.
Use CRISPOR
Go to the CRISPOR website and paste the sequence into the CRISPOR form, select the appropriate off-site genome and PAM type and click Submit.
Export the CRISPOR Results
Once the CRISPOR search is completed, scroll to the bottom of the results and click "Guides all scores".
Choose the location to save the exported results and click Save.
Import the CRISPOR Results
View the target sequence in SnapGene and go Tools → Import Sites from CRISPOR....
In the file picker, choose the CRISPR Excel (.xls) results file and click Import.
SnapGene will parse the Excel file and list the information that has been identified.
Revise the name of the set if required, then click Import CRISPR Sites.
The imported CRISPR sites will be shown in the CRISPR view, and displayed in Map and Sequence view.
Hover over a site to see a tooltip with details for that site.
Save to keep the imported CRISPR sites.
