How do change the default enzymes offered for use in Golden Gate Assembly?
The Default Enzyme list
The SnapGene Golden Gate Assembly tools allow you to choose from an initial default list of 9 Type IIS enzymes that are commonly used for Golden Gate Assembly. The default enzyme is BsaI.
Change the Golden Gate Enzymes List
Click Actions → Golden Gate Assembly → Golden Gate Settings... to show a list of all Golden Gate-compatible Type IIS enzymes in the SnapGene Enzyme database.
Use the check boxes to select the Type IIS enzymes you want to see as listed as options for Golden Gate Assembly.
Use the "Default Golden Gate enzyme" dropdown to specify the enzyme selected when the Golden Gate Assembly tool is started.
The selected enzymes, and the new default will be shown via the "Golden Gate Enzyme" dropdown list when you run Actions → Golden Gate Assembly.
Choose Whether to Automatically Switch to the Golden Gate Enzymes Set
By default, SnapGene will switch the Enzyme set in use to the "Golden Gate Enzymes Set" for all input sequences chosen in the Golden Gate tool.
To retain a sequences specified Enzyme Set, click Actions → Golden Gate Assembly → Golden Gate Settings... and uncheck the option to "Choose the Golden Gate enzymes set..."
The Enzyme Set associated with the input vector will now be retained, but it will be amended if required to include the specified Golden Gate Enzyme.
